Overview

VBI-003 is a **CD47 x EpCAM bispecific monoclonal antibody** being developed by **Virtuoso Therapeutics** for **solid tumors**. Built on a **human IgG1 backbone**, it is designed to bind **CD47** and **EpCAM**, block the **CD47/SIRPα innate immune checkpoint** on EpCAM-positive tumor cells, and preserve IgG1 Fc effector function to promote antitumor immune activity. The bispecific design is intended to improve tumor selectivity by enabling stable binding and SIRPα blockade preferentially where both CD47 and EpCAM are co-expressed. Preclinical data cited for the program include single-agent antitumor activity in xenograft models of **gastric cancer** and **esophageal cancer**, with exploration also reported in **small cell lung cancer** and **colorectal cancer**.
