Overview

VBIT-4 is a small molecule inhibitor of Voltage-Dependent Anion Channel 1 (VDAC1) oligomerization. VDAC1 is a key protein in the outer mitochondrial membrane that regulates the flux of ions and metabolites; its oligomerization is associated with mitochondrial membrane permeabilization and the release of pro-apoptotic factors. While VBIT-4 has demonstrated cytoprotective effects in various disease models (such as diabetes and neurodegeneration) by preventing VDAC1-mediated apoptosis, recent research indicates that its effects may be context-dependent. Specifically, in models of doxorubicin-induced cardiomyopathy, VBIT-4 has been shown to exacerbate mitochondrial dysfunction by increasing mitochondrial iron accumulation and oxidative stress.
