Overview

VCAR33ALLO is an allogeneic, donor-derived chimeric antigen receptor T cell (CAR-T) therapy targeting CD33, a myeloid cell surface antigen highly expressed on the majority of acute myeloid leukemia (AML) blasts. The product is manufactured from healthy lymphocytes obtained from the same donor who provided the patient’s original hematopoietic stem cell transplant. This approach aims to generate CAR-T cells that are HLA-matched to the recipient’s engrafted blood system, potentially improving expansion, persistence, and anti-leukemia activity compared to autologous or off-the-shelf allogeneic products. The therapy is being developed for patients with relapsed or refractory AML after allogeneic hematopoietic cell transplantation (alloHCT), including those previously treated with trem-cel (tremtelectogene empogeditemcel). Clinical development is ongoing in a Phase 1/2 trial (VBP301), evaluating safety and efficacy endpoints such as maximum tolerated dose and clinical response rates[1][2][3][4][5][6][7][8].
