Overview

**VCN-12** is a next-generation oncolytic adenovirus selected from Theriva Biologics' VCN-X discovery program and is derived from their lead clinical product VCN-01 (zabilugene almadenorepvec)[1][2][3][4]. VCN-12 is armed with additional transgenes engineered to improve tumor cell lysis, enhance tumor stroma degradation, and augment antitumor immune responses[1][2][3][4]. The drug is specifically designed as an antitumor agent with potential feasibility for intracranial administration in brain tumors[1][2][3][4]. Preclinical data suggest its mechanism of action may result in significant improvement in antitumor effects compared to earlier generations[1][2][3][4].
