Drug intelligence / Profile preview
Vebicortotamab (also known as becotatug vedotin or MRG003) is an epidermal growth factor receptor (EGFR)-targeted antibody-drug conjugate (ADC) developed by Lepu Biopharma and its subsidiary Shanghai Miracogen. It consists of a humanized IgG1 monoclonal antibody targeting EGFR (becotatug) conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a cleavable valine-citrulline linker. Vebicortotamab binds with high affinity to EGFR on the surface of tumor cells, leading to internalization and subsequent lysosomal protease cleavage of the linker, which releases MMAE to induce cell cycle arrest and apoptosis. In October 2025, it was approved in China under the brand name Meiyouheng (美佑恒) for the treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC) in patients who have failed at least two lines of systemic chemotherapy and PD-1/PD-L1 inhibitor therapy. It is also being evaluated in clinical trials for other EGFR-expressing solid tumors, including biliary tract cancer, head and neck squamous cell carcinoma, and non-small cell lung cancer.
