Vebicorvir is a first-generation, orally available small molecule that acts as a hepatitis B virus (HBV) core protein inhibitor. It was developed to treat chronic HBV infection by targeting the viral core protein, thereby preventing encapsidation of pregenomic RNA and subsequent DNA replication. Vebicorvir suppresses covalently closed circular DNA (cccDNA) formation and has demonstrated effective antiviral activity in clinical studies. Its mechanism of action is orthogonal to standard-of-care nucleos(t)ide reverse transcriptase inhibitors (NrtIs), and it has been studied in combination with these agents for enhanced suppression of HBV[1][2][4][5][8]. The drug was primarily developed by Assembly Biosciences and Antios Therapeutics but its development for hepatitis B has been discontinued[4].
