vebreltinib + andamertinib

Vebreltinib + andamertinib is an oral combination of two small molecule tyrosine kinase inhibitors: vebreltinib (a potent and selective inhibitor of mesenchymal-epithelial transition factor, MET) and andamertinib (PLB1004, an irreversible and highly selective epidermal growth factor receptor [EGFR] tyrosine kinase inhibitor with activity against classical EGFR mutations and exon 20 insertion, and high blood-brain barrier penetration)[2][4]. Developed by Avistone Biotechnology, this dual-target therapy is intended for the treatment of non-small cell lung cancer (NSCLC) patients harboring EGFR mutations who have developed resistance due to MET amplification or overexpression, a known resistance pathway to EGFR-TKI therapy[2][3][4]. The combination is currently in clinical trials, has demonstrated notable efficacy (ORR up to 50-59% in phase Ib/II) and manageable safety, and is being investigated against platinum-based chemotherapy in phase III for advanced/metastatic EGFR-mutated, MET-amplified or overexpressed NSCLC following EGFR-TKI failure[1][2][3][4][5][6].
