The vectorized HER2 antibody is an experimental gene therapy developed by Voyager Therapeutics for the treatment of HER2-positive (HER2+) brain metastases, particularly those originating from breast cancer. This biologic therapy utilizes an adeno-associated virus (AAV) vector, specifically the proprietary TRACER capsid VCAP-102 (a variant of AAV9), to deliver genetic material encoding an anti-HER2 monoclonal antibody. By transducing cells within the central nervous system, the therapy enables the brain to produce therapeutic antibodies locally, thereby overcoming the blood-brain barrier limitations that hinder conventional systemic therapies like trastuzumab. The expressed antibody binds to the Human Epidermal Growth Factor Receptor 2 (HER2), inhibiting tumor cell proliferation and mediating antibody-dependent cell cytotoxicity (ADCC).
