Overview

This is a combination therapy consisting of a VEGF-antisense oligonucleotide, pemetrexed, and cisplatin. The VEGF-antisense oligonucleotide is designed to inhibit the expression of vascular endothelial growth factor A (VEGFA), thereby reducing angiogenesis in tumors by blocking transcription or translation and promoting mRNA degradation via RNase H cleavage. Pemetrexed is an antifolate chemotherapeutic agent that inhibits folate-dependent enzymes involved in nucleotide synthesis, leading to impaired DNA and RNA synthesis and subsequent cell death. Cisplatin is a platinum-based chemotherapy drug that forms DNA crosslinks, resulting in apoptosis of rapidly dividing cells. This combination has been investigated for the treatment of advanced malignant mesothelioma[2][3][8].
