Overview

VEGFA plasmid DNA is a gene therapy approach that involves the delivery of a plasmid, a circular double-stranded DNA molecule, engineered to carry the gene encoding Vascular Endothelial Growth Factor A (VEGFA). The primary goal is to introduce this gene into target cells, enabling them to produce the VEGFA protein. VEGFA is a potent angiogenic cytokine that plays a crucial role in both vasculogenesis and angiogenesis by stimulating the proliferation and migration of vascular endothelial cells, thereby promoting the formation of new blood vessels and improving blood supply to ischemic tissues. This therapeutic strategy is being investigated for conditions characterized by insufficient blood flow, such as peripheral arterial disease and impaired wound healing. While the clinical efficacy of plasmid DNA-based gene therapy can be limited by inefficient transgene expression, ongoing research aims to enhance delivery systems, for instance, through the use of minicircle DNA or co-delivery with other pro-angiogenic factors like FGF2.
