Overview

VEGFR2-169 + gemcitabine is a combination therapy that pairs an immunotherapeutic peptide vaccine with a standard chemotherapy drug. The VEGFR2-169 peptide (elpamotide) targets vascular endothelial growth factor receptor 2 (VEGFR2), which is an essential factor in tumor angiogenesis and pancreatic cancer growth. The peptide vaccine works by inducing cytotoxic T lymphocytes (CTLs) that specifically target VEGFR2, potentially inhibiting tumor angiogenesis. Gemcitabine is a nucleoside analog that inhibits DNA synthesis and is a standard treatment for pancreatic cancer. In clinical trials, this combination has shown promising results with a disease control rate of 67% and median overall survival of 8.7 months in patients with advanced pancreatic cancer. The therapy was well-tolerated at all tested doses (0.5, 1, and 2 mg/body of VEGFR2-169), with no severe adverse effects of grade 4 or higher observed. The peptide vaccine successfully induced specific CTLs in 61% of patients, even when administered alongside gemcitabine[4][6].
