Drug intelligence / Profile preview
VEGFR2-CAR T cells are chimeric antigen receptor (CAR) T cells genetically engineered to target vascular endothelial growth factor receptor 2 (VEGFR2), which is highly expressed on tumor endothelial cells and certain tumor cells. By targeting VEGFR2, these CAR T cells aim to disrupt the tumor vasculature (anti-angiogenic effect) and directly eliminate VEGFR2-expressing tumor cells. Preclinical studies have demonstrated efficacy in models of glioblastoma and other solid tumors, showing improved tumor infiltration and survival. However, clinical translation has been challenging; a Phase I/II clinical trial (NCT01218867) conducted by the National Cancer Institute in patients with metastatic cancers, including metastatic melanoma and renal cell carcinoma, was terminated due to a lack of objective clinical responses.
