Overview

"vehicle + rituximab" refers to a clinical trial intervention arm consisting of the active monoclonal antibody rituximab administered in conjunction with an inactive vehicle or placebo. This combination is frequently utilized as a control group in randomized clinical trials to evaluate the efficacy and safety of a novel investigational drug when added to a background of rituximab standard-of-care. Rituximab is a chimeric IgG1 monoclonal antibody that targets the CD20 antigen, a protein primarily expressed on the surface of B-lymphocytes. Upon binding, rituximab mediates B-cell depletion through antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and the induction of apoptosis. It is a standard-of-care treatment for various B-cell malignancies, such as non-Hodgkin lymphoma and chronic lymphocytic leukemia, as well as autoimmune diseases including rheumatoid arthritis, pemphigus vulgaris, and granulomatosis with polyangiitis.
