Veledimex is an orally bioavailable, small molecule diacylhydrazine-based activator ligand developed as a gene expression regulator for use with the RheoSwitch Therapeutic System (RTS), an ecdysone receptor (EcR)-based inducible gene regulation platform. When administered, veledimex binds to the EcR component of RTS, stabilizing a heterodimeric transcription factor complex that activates transcription from an inducible promoter, thereby enabling controlled expression of target genes such as human interleukin-12 (IL‑12). This system has been investigated primarily in combination with Ad‑RTS‑hIL‑12 for the treatment of recurrent or progressive glioblastoma multiforme and other cancers. Veledimex’s effect is dose-dependent and reversible, allowing precise temporal control over therapeutic gene expression[1][2][4][5][6].
