Overview

Veligrotug is an investigational **intravenous anti-insulin-like growth factor-1 receptor monoclonal antibody** being developed by Viridian Therapeutics for **thyroid eye disease**. It is designed to inhibit IGF-1R signaling, a validated pathogenic pathway in TED, and has been studied in both **active** and **chronic** disease. In Viridian's Phase 3 program, veligrotug used a differentiated **five-infusion IV regimen** and met primary and secondary endpoints in both the THRIVE trial in active TED and THRIVE-2 in chronic TED, with reported benefits on proptosis, diplopia, and clinical activity score. Viridian has also partnered the asset regionally, including a Japan deal with Kissei Pharmaceutical, while positioning veligrotug as the intravenous lead asset in a broader TED franchise that also includes the subcutaneous half-life-extended follow-on antibody VRDN-003.
