Veliparib + bortezomib is an investigational combination therapy consisting of two small molecule drugs with distinct mechanisms of action, primarily explored in oncology. Veliparib (ABT-888) is a poly(ADP-ribose) polymerase (PARP) inhibitor that impairs DNA repair in cancer cells, leading to cell death, especially in tumors with existing DNA repair deficiencies. Bortezomib (marketed as Velcade and Boruzu; code names PS‑341 and MLN‑341) is a reversible inhibitor of the 26S proteasome, disrupting protein degradation pathways within cells and inducing apoptosis by causing accumulation of misfolded or damaged proteins. Bortezomib is approved for the treatment of multiple myeloma and mantle cell lymphoma[2][5][8]. The combination has been studied for potential synergistic effects in various cancers due to their complementary mechanisms—veliparib targeting DNA repair and bortezomib targeting protein homeostasis.
