Veliparib (ABT-888) is an oral small molecule inhibitor of poly(ADP-ribose) polymerase (PARP) enzymes 1 and 2. It is being evaluated in combination with the chemotherapy agents capecitabine (a fluoropyrimidine carbamate) and oxaliplatin (a platinum-based antineoplastic) for the treatment of various advanced solid tumors, including gastrointestinal malignancies like pancreatic and gastric adenocarcinomas, as well as colorectal and ovarian cancers. The combination aims to enhance the cytotoxic effects of chemotherapy by blocking PARP-mediated DNA repair, particularly in tumors with homologous recombination deficiencies or BRCA mutations. This specific regimen has been studied in Phase 1 trials sponsored by the National Cancer Institute (NCI).
