Overview

**veliparib + carboplatin + etoposide** is an investigational combination therapy consisting of three agents: veliparib, an oral small molecule poly(ADP-ribose) polymerase (PARP) inhibitor; carboplatin, a platinum-based alkylating chemotherapeutic; and etoposide, a topoisomerase II inhibitor. The combination is primarily studied for the treatment of extensive-stage small cell lung cancer (ED-SCLC). Veliparib sensitizes cancer cells to DNA-damaging cytotoxic agents (like carboplatin and etoposide) by inhibiting PARP, a key enzyme in DNA repair, thereby enhancing the cytotoxic effects of chemotherapy. This regimen has been evaluated in phase 1 and phase 2 clinical studies in newly diagnosed and previously treated patients with SCLC and other solid tumors. Treatment involves oral administration of veliparib and intravenous delivery of carboplatin and etoposide. The main clinical outcomes demonstrated improved progression-free survival, but no significant increase in overall survival compared to chemotherapy alone. The combination’s safety profile aligns with known effects of the individual components, with hematologic toxicities being the most common adverse events[1][2][3][5].
