This is a combination therapy consisting of three agents: - **Veliparib** (ABT-888): an oral small molecule inhibitor of poly(ADP-ribose) polymerase 1 and 2 (PARP1/2), which impairs DNA repair in cancer cells and potentiates the effects of DNA-damaging agents. - **VX-970** (also known as M6620): a small molecule inhibitor of ataxia telangiectasia and Rad3-related protein kinase (ATR), which is involved in the cellular response to DNA damage. - **Cisplatin**: a platinum-based chemotherapeutic agent that induces DNA cross-linking, leading to apoptosis in rapidly dividing cells. The rationale for this combination is to exploit synthetic lethality by simultaneously inhibiting two key DNA damage response pathways (PARP and ATR) while inducing extensive DNA damage with cisplatin. This approach aims to maximize tumor cell death, particularly in tumors with defects in homologous recombination repair or other vulnerabilities. The triplet has been studied primarily for advanced refractory solid tumors[1][2][4][5].
