Overview
Velneperit (S-2367) is a potent and selective, centrally acting small molecule antagonist of the neuropeptide Y (NPY) Y5 receptor. Discovered and developed by Shionogi, the drug was designed as an oral treatment for obesity. Its mechanism involves blocking the Y5 receptor, a key signaling molecule in the hypothalamus that regulates food intake and energy homeostasis. Although Phase II clinical trials demonstrated statistically significant weight reduction in obese subjects over a one-year period, Shionogi discontinued the program due to results that did not meet the commercial or clinical threshold for further development.
