Veltuzumab + 90Y-epratuzumab tetraxetan is a combination therapy used primarily in the treatment of relapsed or refractory aggressive B-cell non-Hodgkin lymphomas. Veltuzumab is a humanized monoclonal antibody targeting CD20 on B cells, similar to rituximab but with structural and functional differences. 90Y-epratuzumab tetraxetan is a radiolabeled monoclonal antibody that targets CD22 on B cells; it consists of epratuzumab conjugated to the chelator tetraxetan, which binds yttrium-90, delivering targeted beta radiation to malignant B cells. The combination leverages dual targeting of two distinct B-cell antigens (CD20 and CD22) for enhanced efficacy without increased toxicity compared to monotherapy. Veltuzumab depletes peripheral blood B-cells prior to administration of the radioimmunotherapy agent, while 90Y-epratuzumab delivers cytotoxic radiation directly into tumor cells via internalization after binding CD22. This regimen has shown encouraging activity and manageable safety in patients with aggressive lymphoma who have failed standard therapies[1][2][3][4]. Both agents were originally developed by Immunomedics.
