Overview

Vemurafenib + trientine is an investigational combination therapy being studied for the treatment of BRAF-mutated metastatic melanoma. Vemurafenib is a small molecule inhibitor targeting the BRAF V600E mutation and is approved for use in unresectable or metastatic melanoma with this mutation. It works by inhibiting the activity of mutant BRAF kinase, thereby blocking downstream MAPK/ERK signaling and reducing tumor cell proliferation. Trientine (triethylenetetramine) is a copper chelator approved for Wilson's disease; it binds excess copper to facilitate its excretion from the body. In this combination regimen, trientine’s role as a copper chelator may disrupt cellular signaling pathways that require copper as a cofactor—potentially enhancing anti-tumor effects when used alongside vemurafenib[1][4]. The combination remains investigational in oncology.
