Overview

Venanprubart is an investigational human IgG4 kappa monoclonal antibody that acts as an agonist of the inhibitory checkpoint receptor B and T lymphocyte attenuator (BTLA), being developed primarily for autoimmune diseases such as systemic lupus erythematosus, chronic plaque psoriasis, and primary Sjögren’s syndrome.[1][5][6][11] By stimulating BTLA signaling on lymphocytes, venanprubart is intended to dampen aberrant immune activation and inflammation, functioning as an anti‑inflammatory and immunomodulatory biologic rather than a classic immunostimulatory checkpoint inhibitor.[1][6] The antibody, originally associated with Sanford Burnham Prebys Medical Discovery Institute and subsequently developed by Eli Lilly, has reached phase 2 development in systemic lupus erythematosus, with earlier-phase work in psoriasis and exploratory work in primary Sjögren’s syndrome, but recent reports indicate discontinuation or lack of ongoing development in some indications.[1][3][6][11]
