**venetoclax + daratumumab + dexamethasone** is an investigational, three-drug combination therapy evaluated primarily in relapsed or refractory multiple myeloma, particularly in patients with the t(11;14) chromosomal translocation. - **Venetoclax** is a small molecule, orally available selective inhibitor of B-cell lymphoma 2 (BCL-2), inducing apoptosis in BCL-2 dependent cancer cells. - **Daratumumab** is a monoclonal antibody targeting CD38 on myeloma cells, inducing cell death through antibody-dependent cellular cytotoxicity, complement-dependent cytotoxicity, phagocytosis, and immune modulation (including reduction of regulatory T-cells and activation of effector T-cells). - **Dexamethasone** is a synthetic glucocorticoid corticosteroid used for its anti-inflammatory and immunosuppressive effects and as an antineoplastic agent in hematologic malignancies. The regimen has shown high overall response rates and deep remissions (including minimal residual disease negativity) in heavily pretreated multiple myeloma, especially those with t(11;14), and is under active investigation in early phase clinical trials[1][3][7][9].
