This is a four-drug combination regimen consisting of venetoclax, dexamethasone, ixazomib, and pomalidomide. Venetoclax is a selective BCL2 inhibitor that induces apoptosis in cancer cells by blocking the anti-apoptotic protein BCL2. Dexamethasone is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressant properties. Ixazomib is an oral proteasome inhibitor that selectively inhibits the 20S proteasome subunit beta type 5, disrupting protein degradation and promoting apoptosis in myeloma cells[6]. Pomalidomide is an immunomodulatory agent that enhances immune response against tumor cells and has direct antineoplastic effects. This combination targets multiple pathways involved in plasma cell survival and proliferation, aiming to improve outcomes for patients with relapsed or refractory multiple myeloma or AL amyloidosis[1][6][7].
