Overview

This investigational combination therapy consists of venetoclax, everolimus, and Enrylaze (asparaginase erwinia chrysanthemi (recombinant)-rywn). It is being evaluated in the ALL-TARGET trial, a Phase 2 precision medicine study led by Philippe Rousselot and the Group for Research on Adult Acute Lymphoblastic Leukemia (GRAALL). The regimen is designed for patients aged 15 and older with relapsed or refractory T-cell acute lymphoblastic leukemia (T-ALL). The combination targets three critical signaling pathways: BCL-2 (inhibited by venetoclax), mTOR (inhibited by everolimus), and asparagine metabolism (depleted by Enrylaze). Treatment selection in the trial is guided by functional companion drug testing to identify individual patient signaling alterations.
