The Venezuelan equine encephalitis virus (VEEV) DNA vaccine is a prophylactic genetic immunization designed to protect against infection by the mosquito-borne alphavirus VEEV. The most studied constructs encode the codon-optimized envelope glycoprotein genes or the full 26S structural gene region (C-E3-E2-6K-E1) of the IAB strain Trinidad donkey of VEEV. Delivered via intramuscular electroporation or jet injection, this DNA vaccine induces robust humoral and cellular immune responses in animal models and has advanced to Phase 1 human clinical trials. It elicits strong neutralizing antibody titers and Th1-type cellular immunity, providing complete protection from lethal aerosol challenge in mice and significant protection in non-human primates. The mechanism of action is based on expression of viral antigens from transfected host cells leading to an adaptive immune response targeting VEEV[1][3][4][5][6].
