VENT-03

VENT-03 is a first-in-class, orally administered small molecule inhibitor of cyclic GMP-AMP synthase (cGAS), developed by Ventus Therapeutics. cGAS is an intracellular pattern recognition receptor that detects cytoplasmic double-stranded DNA, leading to the activation of inflammatory pathways implicated in autoimmune and cardiometabolic diseases. By selectively inhibiting cGAS, VENT-03 suppresses DNA-driven type I interferon activity and NF-kB-mediated pro-inflammatory cytokine production, thereby reducing systemic inflammation and modulating adaptive immune responses. Preclinical studies demonstrated robust pharmacodynamic activity and efficacy in models of auto-inflammation. In Phase 1 clinical trials, VENT-03 was safe, well-tolerated at all tested doses, showed full target inhibition, and enabled once-daily dosing due to favorable pharmacokinetics. The drug is advancing into Phase 2 development for systemic lupus erythematosus (SLE) with potential applications across a range of autoimmune disorders and cardiometabolic diseases[1][2][3][4][5][6].
