Overview

Vereirumab (formerly UPB-101) is a human IgG1 kappa monoclonal antibody that targets the Cytokine Receptor-Like Factor 2 (CRLF2), also known as the Thymic Stromal Lymphopoietin receptor (TSLPR). Developed by Upstream Bio, it is currently in Phase 2 clinical development for the treatment of severe asthma and chronic rhinosinusitis with nasal polyps (CRSwNP). By binding to CRLF2, vereirumab blocks the signaling of TSLP, an upstream "alarmin" cytokine that triggers multiple downstream inflammatory pathways, including Th2 and Th17 responses. Unlike tezepelumab, which targets the TSLP ligand, vereirumab targets the receptor itself, which may provide more complete or sustained inhibition of the pathway. The drug is designed for subcutaneous administration and aims to provide a long-acting therapeutic option for patients with eosinophilic and non-eosinophilic inflammatory profiles.
