Overview

Verubulin is a small molecule quinazoline derivative with antineoplastic activity. It acts as a microtubule destabilizer by binding to β-tubulin and inhibiting tubulin polymerization, thereby disrupting microtubule formation. This leads to mitotic spindle assembly disruption, cell cycle arrest in the G2/M phase, and subsequent cell death. Verubulin also functions as a vascular disrupting agent (VDA), selectively targeting tumor vasculature and causing acute ischemia and massive tumor cell death. Notably, it is not a substrate for several multidrug resistance ABC transporters and can cross the blood-brain barrier, accumulating in brain tissue. Verubulin has been investigated primarily for recurrent glioblastoma (GBM), brain cancer, lung cancer, melanoma, and other solid tumors[1][2][5][6][8].
