Overview

**Verubulin + temozolomide** is a combination regimen under investigation for the treatment of newly diagnosed glioblastoma multiforme (GBM). Verubulin is a microtubule destabilizer and vascular disrupting agent that has demonstrated high brain penetration and disrupts tumor vasculature by inhibiting microtubule polymerization. Temozolomide is an alkylating agent that methylates DNA at the O6 and N7 positions of guanine, leading to apoptosis in tumor cells, especially those with impaired DNA repair. The combination aims to leverage the synergistic effects of two distinct mechanisms—microtubule disruption and DNA alkylation—to enhance tumor cell death and overcome resistance to temozolomide. This combination is currently being evaluated in clinical trials for glioblastoma where both drugs are administered together with radiation therapy[3][4][5].
