Vesatolimod is an orally administered small molecule that acts as a selective agonist of toll-like receptor 7 (TLR7). By binding to and activating TLR7, which is primarily expressed on plasmacytoid dendritic cells and B cells, vesatolimod stimulates the innate immune system. This activation leads to the induction of interferon-stimulated gene expression and cytokine production (including type I interferon-alpha), as well as activation of natural killer cells, phagocytic cells, plasmacytoid dendritic cells, and T cells. Vesatolimod has been investigated primarily for its potential role in HIV cure strategies—aiming to reduce viral reservoirs during antiretroviral therapy—and was also studied for hepatitis B virus infection. The drug is being developed by Gilead Sciences in collaboration with Aelix Therapeutics and the National Institute of Allergy and Infectious Diseases[1][2][5].
