Overview

Vesencumab (MNRP1685A) is a humanized IgG1 monoclonal antibody designed to target neuropilin-1 (NRP1), a transmembrane glycoprotein that serves as a co-receptor for vascular endothelial growth factor (VEGF). By binding to the b1b2 domain of NRP1, vesencumab prevents the interaction between VEGF-A165 and NRP1, thereby inhibiting the potentiation of VEGF receptor-2 (VEGFR2) signaling and disrupting pro-angiogenic pathways in the tumor microenvironment. Developed by Genentech (Roche), the drug was investigated in Phase I clinical trials for the treatment of advanced or metastatic solid tumors, both as a monotherapy and in combination with bevacizumab. However, clinical development was discontinued due to safety concerns, specifically a high incidence of immune-mediated thrombocytopenia observed in treated patients.
