The VesiculoVax Trivalent Filovirus Vaccine is a recombinant viral vector vaccine candidate designed to provide broad protection against the three primary causes of filoviral hemorrhagic fever: Ebola virus (Zaire), Sudan virus, and Marburg virus. Developed using the VesiculoVax platform, originally by Profectus BioSciences and later advanced by the Sabin Vaccine Institute, the vaccine is a blend of three separate vectors. Each vector is a live-attenuated recombinant vesicular stomatitis virus (rVSV) engineered to express the surface glycoprotein (GP) of one of the target filoviruses. The platform incorporates specific safety modifications, including the translocation of the VSV N gene (N4) and the truncation of the G protein cytoplasmic tail (CT1), which significantly reduces neurovirulence while maintaining high immunogenicity. The vaccine works by inducing potent neutralizing antibodies and cellular immune responses against the viral glycoproteins, providing both rapid and durable protection, as demonstrated in non-human primate models.
