Overview

Vespula maculifrons venom protein refers to the allergenic components derived from the venom of the Eastern Yellowjacket, used clinically for both the diagnosis and treatment of Hymenoptera venom allergy. As a diagnostic agent, it is employed in skin prick or intradermal testing to identify IgE-mediated sensitization in patients who have experienced systemic reactions to insect stings. As a therapeutic agent in venom immunotherapy (VIT), the extract is administered in escalating doses to induce immunological tolerance. The mechanism of action involves a shift from a Th2-biased allergic immune response to a Th1/Treg-mediated response, characterized by the production of interleukin-10 (IL-10) and the induction of allergen-specific IgG4 "blocking" antibodies. These IgG4 antibodies compete with IgE for allergen binding, thereby preventing the cross-linking of IgE on mast cells and basophils and inhibiting the release of inflammatory mediators like histamine and leukotrienes.
