Overview

Vespula squamosa venom protein, commonly known as Southern Yellowjacket venom extract, is a biological product used for the diagnosis and treatment of Hymenoptera venom allergy. It is primarily indicated for venom immunotherapy (VIT) to reduce the risk of systemic allergic reactions, including anaphylaxis, in individuals with a history of severe sensitivity to Southern Yellowjacket stings. The extract contains major allergens such as Phospholipase A1 (Vesp s 1), Hyaluronidase (Vesp s 2), and Antigen 5 (Vesp s 5). The mechanism of action involves desensitization of the immune system, characterized by a shift from a Th2-dominated allergic response to a Th1 and regulatory T-cell (Treg) response. This modulation leads to the production of allergen-specific IgG4 antibodies that act as blocking antibodies, neutralizing the venom allergens before they can bind to IgE on mast cells and basophils, thereby preventing the release of inflammatory mediators.
