Overview
Vestipitant + paroxetine is a combination of two small molecule drugs investigated primarily for the treatment of chronic tinnitus. Vestipitant is a potent and selective neurokinin 1 (NK1) receptor antagonist developed by GlaxoSmithKline, with anxiolytic and antiemetic properties, as well as potential utility in insomnia and anxiety disorders. Paroxetine is a selective serotonin reuptake inhibitor (SSRI) widely used to treat depression, anxiety disorders, obsessive-compulsive disorder, posttraumatic stress disorder, and menopausal hot flashes. The combination was studied to assess whether dual modulation of NK1 receptors and serotonin reuptake could provide benefit in tinnitus; however, clinical trials found that the combination was not effective for this indication[1][3][7].
