Overview
**Vestipitant + vofopitant** is a combination of two investigational small molecule drugs, both of which are highly selective antagonists of the neurokinin-1 (NK1) receptor. This receptor modulates the action of substance P, a neuropeptide involved in emesis (vomiting), anxiety, and pain. Both vestipitant and vofopitant have been studied individually and in combination for disorders such as chronic tinnitus, sleep disturbances, and the prevention of nausea and vomiting, though they have not demonstrated consistent efficacy in phase 2 testing. Developed predominantly by GlaxoSmithKline, the combination targets central NK1-mediated pathways suggested to play a role in central nervous system disorders[1][4][5].
