Vexo-AAV6-iCasp9 is a gene therapy product consisting of an exosome-associated adeno-associated virus serotype 6 (AAV6) vector carrying an inducible caspase 9 (iCasp9) suicide gene. The iCasp9 transgene is activated in the presence of the dimerizer drug AP20187, triggering apoptosis selectively in transduced cells. Vexo-AAV6-iCasp9 has demonstrated enhanced cytotoxicity and tumor regression in preclinical models of hepatocellular carcinoma and breast cancer. The exosome-based (vexosome) delivery adds functional efficiency and ease of isolation for clinical use. This approach is designed for intratumoral gene therapy targeting solid malignancies through induction of programmed cell death[1][2][3].
