The combination of **VG161 + nivolumab** consists of VG161, a genetically engineered oncolytic herpes simplex virus type 1 (HSV-1) carrying multiple antitumor immunomodulating genes (interleukin-12, interleukin-15, IL-15 receptor alpha, and a PD-L1 blocking peptide), and nivolumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-1. VG161 acts to directly lyse tumor cells and modify the tumor microenvironment by stimulating both innate and adaptive immune responses, while its engineered payloads further augment immune activation locally. Nivolumab blocks the PD-1 receptor on T cells, releasing their inhibition and enhancing systemic antitumor immune activity. This combination is being investigated primarily in advanced solid tumors, such as hepatocellular carcinoma, pancreatic cancer, and gastric/gastroesophageal junction adenocarcinoma, aiming to synergistically overcome tumor immune evasion and produce sustained antitumor effects[1][2][3][4][6].
