Overview

VG201 is a first-in-class oncolytic virus therapy developed by Virogin Biotech. It is based on a transcriptional and translational dual-regulated (TTDR) herpes simplex virus type 1 (HSV-1) backbone. VG201 is engineered to enhance oncolytic activity specifically in tumors expressing carcinoembryonic antigen (CEA). The virus encodes immune-stimulating cytokines interleukin 12 and interleukin 15/IL-15Rα, which are designed to modulate the tumor microenvironment and boost both innate and adaptive anti-tumor immunity. Preclinical studies suggest that VG201 has greater potency than non-attenuated oncolytic viruses with an acceptable safety profile, and it has shown synergistic effects when combined with anti-PD-1 monoclonal antibodies or targeted therapies. Clinical development includes Phase 1 trials for advanced solid tumors, using both intratumoral injection and intravenous administration[1][6][8].
