VG2025 is a recombinant oncolytic herpes simplex virus type 1 (HSV-1) engineered for cancer therapy. It utilizes transcriptional and translational dual regulation (TTDR) of key viral genes to enhance safety and promote tumor-specific replication without reducing virulence. The essential HSV-1 immediate-early protein ICP27 is controlled by a tumor-specific carcinoembryonic antigen (CEA) promoter, while the neurovirulence factor ICP34.5 is regulated via multiple microRNA-binding sites. VG2025 also incorporates interleukin-12 (IL-12), interleukin-15 (IL-15), and IL-15 receptor alpha subunit as payloads, which are under the control of a tumor promoter CxCR4, to boost antitumor immune responses. The drug is administered via intratumoral injection in patients with advanced solid tumors that have progressed after standard therapies[1][2][3][7].
