VG9-IL-24 is a recombinant oncolytic vaccinia virus (strain Guang9) engineered to express the human interleukin-24 (IL-24) gene. Its mechanism combines selective replication in tumor cells (leading to direct oncolysis) and localized overexpression of IL-24, a tumor suppressor cytokine. IL-24 induces apoptosis, G2/M cell cycle arrest, and inhibits cell proliferation specifically in cancer cells, largely sparing normal cells[1][2][3][4]. VG9-IL-24 reduces levels of PI3K, inhibits AKT phosphorylation, activates GSK-3β, and promotes β-catenin degradation, leading to apoptotic effects[1]. It has demonstrated potent antitumor effects in xenograft models of breast and colorectal cancers, both by direct killing and by stimulating antitumor immune responses, including enhanced secretion of IFN-γ, IL-6, TNF-α, and IL-4, and “bystander” activity against neighboring non-injected tumors[2][3][4].
