VGX-100 is a fully human IgG1λ monoclonal antibody that specifically binds to and neutralizes vascular endothelial growth factor C (VEGF-C). By inhibiting VEGF-C, VGX-100 prevents its interaction with the VEGFR2 (KDR/FLK1) and VEGFR3 (FLT4) receptors, thereby blocking downstream signaling pathways involved in angiogenesis and lymphangiogenesis. This mechanism may inhibit the proliferation of vascular and lymphatic endothelial cells, potentially reducing tumor growth, metastasis, and spread. Developed primarily for oncology indications such as advanced solid tumors, VGX-100 has also shown preclinical potential in treating dry eye disease by reducing inflammation and corneal tissue damage. The drug was initially developed by Vegenics (a subsidiary of Circadian Technologies), with clinical trials conducted for various late-stage cancers both as monotherapy and in combination with bevacizumab[1][2][4][5][6].
