vGY2-IL-15sa is a novel TROP2-targeted immunocytokine fusion protein designed for cancer immunotherapy. It consists of a shark-derived Variable New Antigen Receptor (VNAR), specifically the high-affinity and low-internalizing vGY2, fused to an interleukin-15 (IL-15) super-agonist complex. The super-agonist component is composed of the IL-15N72D mutation and the IL-15 receptor alpha (IL-15Rα) sushi domain. By targeting Trophoblast cell surface antigen 2 (TROP2), which is frequently overexpressed in solid tumors such as non-small cell lung cancer (NSCLC), the fusion protein localizes IL-15 signaling to the tumor microenvironment. This approach activates cytotoxic T cells and natural killer (NK) cells while minimizing the systemic toxicity typically associated with IL-15 therapy. The use of a low-internalizing VNAR ensures that the IL-15 signaling remains active on the tumor cell surface to maximize the stimulation of neighboring immune cells.
