VH3810109 is a next-generation broadly neutralizing antibody (bNAb) targeting the CD4 binding site on the HIV envelope glycoprotein gp120. It is a human IgG1 monoclonal antibody of the VRC01 class and is a modified version of the parent bNAb N6, engineered with an LS mutation to extend plasma half-life. VH3810109 demonstrates broad and potent neutralizing activity against 98% of tested HIV isolates, including those resistant to VRC01. Its mechanism involves binding to gp120, thereby inhibiting viral entry into host cells and potentially inducing immune-mediated clearance via Fc receptor interactions such as phagocytosis or antibody-dependent cellular cytotoxicity (ADCC). The drug is being developed for both prevention and treatment/cure strategies in HIV infection. Clinical studies have shown that intravenous administration can produce substantial reductions in viral load in treatment-naive participants; subcutaneous dosing results in lower exposure and antiviral response compared to intravenous dosing. The drug has been well tolerated with no serious adverse events reported[1][2][4][7].
