Overview

VHB937 is a fully human monoclonal antibody that acts as a selective agonist and stabilizer of TREM2 (Triggering Receptor Expressed on Myeloid Cells 2), a receptor expressed on microglia in the central nervous system. By activating TREM2, VHB937 increases its cell surface expression by reducing shedding, enhances Syk-phosphorylation, and drives calcium signaling within microglia. This activation promotes key microglial functions such as chemotaxis and phagocytosis, supporting neuronal health by reducing neuroinflammation and protecting against neurodegeneration. Preclinical studies have shown that VHB937 can induce a neuroprotective phenotype in microglia, reduce dystrophic neurites, ameliorate astrogliosis, and decrease pro-inflammatory markers both *in vitro* and *in vivo*. The drug is being developed primarily for the treatment of amyotrophic lateral sclerosis (ALS) and potentially other neurodegenerative diseases.
