VHC-001 is an investigational immunotherapy consisting of a complex of recombinant human interleukin-2 (IL-2) and a humanized IgG1 monoclonal antibody directed against IL-2. Developed by the Vall d'Hebron Institute of Oncology (VHIO), this "immunocomplex" approach is designed to selectively direct IL-2 signaling toward effector T cells (CD8+) and natural killer (NK) cells while minimizing the activation of regulatory T cells (Tregs) and reducing systemic toxicities like vascular leak syndrome. By binding to a specific epitope on IL-2, the antibody prevents IL-2 from interacting with the high-affinity IL-2 receptor alpha (CD25), which is constitutively expressed on Tregs and vascular endothelial cells, while maintaining its ability to bind the intermediate-affinity IL-2 receptor beta/gamma (CD122/CD132) on effector cells. It is being evaluated for the treatment of solid tumors, including cutaneous melanoma, non-small cell lung cancer (NSCLC), and cervical cancer.
