The VHH-based KappaLink platform is a preclinical-stage therapeutic technology developed by Cerberus Therapeutics. It utilizes single-domain antibodies (VHHs or nanobodies) engineered to form an adduct that binds the immunoglobulin kappa (Igκ) light chain present on all naturally circulating immunoglobulins (including IgG, IgM, IgA, and IgE). This binding brings together infected cells and a diverse set of Fc receptor–positive effector cells. The result is rapid and effective activation of both B cell and T cell immune responses. The primary aim of the platform is to treat and prevent infections by leveraging endogenous antibody pools for broad immune engagement[7][9]. VHHs are derived from camelid heavy-chain-only antibodies; they are small, robust antigen-binding fragments with high stability and versatility in engineering[1][2]. The modularity of the KappaLink approach allows for potential applications across infectious diseases where enhanced immune recruitment is beneficial.
