Vi-CRM197

Vi-CRM197 is a monovalent typhoid conjugate vaccine (TCV) originally developed by the Novartis Vaccines Institute for Global Health (NVGH) and later licensed to Biological E. Limited. It consists of the Vi capsular polysaccharide of *Salmonella enterica* serovar Typhi (purified from *Citrobacter freundii*) chemically conjugated to CRM197, a non-toxic mutant of diphtheria toxin that serves as a carrier protein. This conjugation converts the T-cell independent Vi polysaccharide into a T-cell dependent antigen, enabling the induction of immunological memory and robust antibody responses in infants and young children. The vaccine has been evaluated in multiple clinical trials, including a landmark human challenge study at the University of Oxford, and is commercialized under the brand name Typhibev.
